There are three main types of raw materials for the production of plant capsules on the market.

One is hypromellose (HPMC), extracted from cellulose raw materials and is one of the commonly used pharmaceutical excipients widely used worldwide. Of course, when used in capsule preparation, some gelling agents such as carrageenan usually need to be added.

The second is pullulan, a water-soluble polysaccharide produced through fermentation, and regulatory agencies in various countries have widely recognized its safety. It has been used in commercial production for more than 25 years and is commonly used in the food and drug industry. At present, pullulan is produced by Aureobasidium pullulans using starch hydrolysate, sucrose, or other sugars as substrates.

The third is vegetable starch, which mainly refers to hydroxypropyl starch, which has stable chemical properties, a wide range of sources, and a large amount. Compared with hypromellose and pullulan capsules, the raw material cost should be much lower, but there are more significant challenges in the preparation process.

The amino acids in gelatin may cause cross-linking reactions, leading to disintegration of the drug or reduced dissolution performance. The gelatin used is obtained chiefly from the hydrolysis of materials such as the skin and bones of pigs and cattle. To solve the above problems, the industry has conducted a lot of research and found various alternative materials for gelatin. Among them, pullulan is an extracellular water-soluble mucilage polysaccharide fermented from corn as a raw material. It has good film-forming properties and is extremely low. With high oxygen permeability, the made hard shell capsules have a uniform and transparent film formation and are considered the most likely new material to replace gelatin.
